Lantern Pharma (NASDAQ: LTRN), a clinical-stage, AI-driven precision oncology company, has received regulatory and Institutional Review Board approvals in Taiwan to open Part 2 of its Phase 2 HARMONIC trial of LP-300, a potential treatment for non-small cell lung cancer patients with the EGFR exon 21 L858R mutation. The lead Taiwan site is now open to accrual and actively screening patients, with two additional sites expected to receive IRB approval in the fourth quarter. In the United States, a central IRB has approved the amended protocol, and clinical sites have begun identifying eligible patients. These developments mark a significant step forward for a trial targeting a patient subgroup that has historically shown poorer responses to standard tyrosine kinase inhibitor therapies, potentially addressing a critical gap in lung cancer treatment.
The L858R mutation accounts for roughly 40% of EGFR mutations in lung cancer and is associated with inferior outcomes compared to other EGFR mutations, such as exon 19 deletions. Patients with this mutation often experience disease progression after tyrosine kinase inhibitors, leaving limited effective options. The HARMONIC trial's Part 2 will enroll patients with the L858R mutation who have progressed after TKI treatment and will allow LP-300 to be administered for up to eight cycles in combination with carboplatin and pemetrexed, up from six cycles previously. This expanded dosing schedule could potentially deepen and prolong treatment responses, a key goal in a disease where durable remissions are rare. Preliminary data from the trial showed a median progression-free survival of 8.9 months among nine L858R patients who completed six cycles, compared with 8.3 months among all 16 L858R patients evaluated. While the company cautions that these findings are preliminary, exploratory, and based on small patient numbers, the trend suggests that LP-300 may offer added benefit for this difficult-to-treat population.
Lantern's approach leverages its proprietary RADR artificial intelligence and machine-learning platform, which integrates large-scale genomic and biological data to identify patients most likely to benefit from its therapies. By focusing on biomarker-driven clinical development, the company aims to reduce the cost, pace, and risk of oncology drug development. The Taiwan approvals not only expand the trial's geographic reach but also validate the company's strategy of using AI to guide precision oncology. If successful, LP-300 could become a much-needed option for L858R patients, potentially improving survival outcomes and quality of life. For investors, the news signals tangible progress in a pipeline that includes LP-184, LP-284, and a CNS-focused subsidiary, Starlight Therapeutics.
